The culture cells are first centrifuged in combination with ice-cold buffer. After this, it is disrupted to release the cell nuclei. Next step of this process includes spinning the cell mixture so that cytoplasmic proteins can be released. Then, the cell mix is kept back in the ice-cold buffer and centrifuged again.  This step helps in releasing of nuclear proteins. The last step of this protein extraction method includes the utilization of warm buffer for releasing cytoskeleton proteins.

In this method, protein samples are blended with liquid nitrogen to make them brittle. After this, the sample is grind using a pestle. This process of grinding ensures low risk to the protein’s integrity.

The basic or first step of this method is chopping of the tissue and homogenizing it before initiating the lysis process. Tissue surface gets the surface to break down due to mechanical disturbances. Centrifugation step is done to make the proteins available.
