Extraction of proteins from Culture Cells Or Centrifugation Method
This method is performed by making use of cells as they only need lysis of the membrane for extracting the proteins. The main part of this method is that it reduces some steps and saves time. The culture cells are first centrifuged in combination with ice-cold buffer. After this, it is disrupted to release the cell nuclei. Next step of this process includes spinning the cell mixture so that cytoplasmic proteins can be released. Then, the cell mix is kept back in the ice-cold buffer and centrifuged again. This step helps in releasing of nuclear proteins. The last step of this protein extraction method includes the utilization of warm buffer for releasing cytoskeleton proteins.
Liquid Nitrogen Grinding method of extraction
In this method, protein samples are blended with liquid nitrogen to make them brittle. After this, the sample is grind using a pestle. This process of grinding ensures low risk to the protein’s integrity.
